I skinned my door with 2" rigid foam and then 1/2" plywood over that, both on the inside. Then weather stripping around the entire opening. I made the door so that it barely cleared the threshold.
The Coolbot website says never use fiberglass insulation because condensing moisture will make the insulation wet in the wall and that will get funky and lose efficiency.
Additionally, if your room has a cement floor you will want to insulate that to reduce sweating. Keeping humidity low will help prevent frosting over of the Ac unit.  I have a dehumidifier running in mine full time here in Georgia. I have to empty the thing every 2-3 days in the summer. I have a slab in mine and per Coolbot recommendations I laid 2" rigid foam on the floor and then fit 3/4" t&g plywood over that with construction adhesive in the interfacing t&g, cutting it skin tight to the walls. So tight I had to jump up and down on it to seat it down. No fasteners.
This approach makes the door close up against almost 3" of the insulated floor. I put a strip of weatherstripping on that face of the floor.  39 to 42 degrees depending on what I have in there for 26 months now.
Harvesting on Monday or Tuesday or Wednesday and holding till Saturday is no issue. I love this thing.
