Two days is a little much, but I recommend an overnight soak at least, especially if the cuttings have been in storage a while. The cuttings seem able to generate callous more easily when they have a full load of water in the cells. I wrote this up in The Grape Grower.

I was talking to a friend the other day who had just pruned his grapes. He was told to soak them in water for 2 days before rooting them. Has anyone else heard of this? Would it's purpose be to encourage callousing?
